When he was five years old, his family moved to Australia. Mesure graduated from the National Academy of Dramatic Art in 1995. He then moved to New Zealand and started performing.

He has written, directed, designed and performed over 30 productions for the University of Sydney Drama Society. In 1995, his first television appearance was in the Mirror, Mirror series. From 1996 to 1998, Mesure played Ryan Walters on the City Life series. The role earned him a New Zealand Television Award nomination for Best Actor. From 1997 to 2001, Mesure starred in the Xena: Warrior Princess series. From 2000 to 2003, he played Kees Van Dam in the TV series “Street Law” for which he won the New Zealand Film and Television Awards for Best Supporting Actor. From 2005 to 2007, Mesure played JD Pollack on the Crossing Jordan franchise. From 2009 to 2010, he played Zane Gerard in the Outrageous Fortune series, and in 2010 played Alex Ross in This Is Not My Life. Charles played Kyle Hobbs in the TV series “V” and Ben Faulkner in the “Desperate Housewives” series.
